Structure of GHK-Cu

  • Histidine: A (semi-)essential amino acid required for histamine production, tissue repair, and protein synthesis.
  • Lysine: An essential amino acid for proteins that the body cannot produce. Its key characteristic is its solubility in water, and therefore also in tissues, due to its positive charge. Together, these three amino acids form a chelate, derived from the Greek word “chele,” meaning “claw,” which firmly but reversibly holds the copper ion.

GHK-Cu and aging

GHK-Cu is an endogenous compound first isolated from human plasma in 1973. A very interesting observation is that the amount of this peptide in blood and tissues is age-dependent. The concentration is highest in young adults; a 20-year-old has an average concentration of 200 ng/ml. With age, this concentration decreases, with a 60-year-old having an average concentration of 80 ng/ml. This observation raises the question: what is the role of this peptide in the body, and why does its concentration decrease with age?

Properties of GHK-Cu

Several properties of GHK-Cu can be described as those of a signaling molecule and gene regulator. The copper in the complex is not merely passive; it is an essential cofactor that can facilitate interactions with specific cell receptors or DNA sequences. According to various animal and in-vitro (cell culture) studies, GHK-Cu appears to be involved in several molecular interactions. Among other things, it appears to bind to specific regions on the DNA, which can lead to changes in gene expression. In addition, it has been associated with modulating the activity of genes involved in a diverse spectrum of cellular processes, including:

  • Cell migration and differentiation: It can influence the movement and specialization of cells such as fibroblasts and keratinocytes.
  • Synthesis of extracellular matrix components: It is associated with genes for proteins such as collagen (types I, III, and IV), elastin, decorin, and fibronectin. These are the structural scaffolding of the skin and other tissues.
  • Modulation of enzyme activity: It can interact with enzymes such as collagenase and elastase, which are involved in the breakdown of collagen and elastin.
  • Interaction with growth factors: It appears to work synergistically or modulate with certain endogenous growth factors.

At the cellular level, its behavior is often described as “normalizing.” It appears to be able to return cells in a deregulated or stressed state to a more homeostatic, healthy function. This has been observed in cells exposed to oxidative stress or damaging agents.
